Vitamin A

Single-ingredient vitamin A reference formula at 3 mg per serving, developed as a softgel or capsule, with IU and mcg RAE label options listed separately.

Vitamin A is a reference formula for brands building a single-ingredient vitamin A product around the three ways that reference labels state the amount: milligrams (3 and 7.5 mg), international units (10,000 and 25,000 IU) and micrograms of retinol activity equivalents (3,000 and 7,500 mcg RAE). The default amount is 3 mg of vitamin A per serving, and the development work centers on the source form (retinyl ester or beta-carotene), the unit used on the label, and the amount tier for each sales market.

Softgel is the lead format because vitamin A is fat soluble and is carried in an oil fill, while a capsule version uses an oil-based or dry vitamin A beadlet. The units are listed apart and are not converted into one another, because the factor depends on which form of vitamin A the label declares.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
Vitamin A3 mg (default amount)Retinyl palmitate in an oil solution, specified as vitamin A (retinol) content; 3 mg vitamin A per servingRetinyl acetate, or beta-carotene as a provitamin A source with its own declared basis and separate specification

Amounts are stated as mg of vitamin A as retinol (C20H30O, 286.5 g/mol; PubChem CID 445354); 3 mg equals 3,000 mcg. Retinyl palmitate (C36H60O2, 524.9 g/mol; CID 5280531) carries 54.6% retinol by weight, so 3 mg of retinol needs about 5.5 mg of the pure ester; retinyl acetate (C22H32O2, 328.5 g/mol; CID 638034) carries 87.2%. Reference labels that state IU or mcg RAE are kept as separate option sets, and the supplier assay and the declared basis set the input.

Intended Benefits & Formula Rationale

This formula is designed for products positioned around daily vitamin A intake and eye nutrition. Vitamin A is included as the single active so that the source form, the amount and the unit can be set without interaction with other actives.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Daily vitamin A intakeVitamin AIncluded as a group of vital micronutrients, with retinyl esters from animal products and provitamin A carotenoids from vegetables and fruits as the dietary forms [1]. Evidence: Referenced
Visual nutritionVitamin APositioned around eye nutrition because the body converts vitamin A and provitamin A into active forms such as retinal that interact with opsin in the retina [1]. Evidence: Referenced

One source form and one unit keep the specification traceable. The reference amounts range from 3 to 7.5 mg and from 10,000 to 25,000 IU, so the amount, the unit, and the label statement are decisions for the brand and its market review, taken together with the safety points below.

Available Formats

The vitamin A reference formula is built for a softgel version and a capsule version; counts per serving are design choices that the brand can adjust.

Raw Material Input & Fill (Design Estimate)

Design estimates. The input is the retinol amount divided by the retinol content of the ester at an assumed 100% assay of the pure ester: retinyl palmitate at 54.6% gives about 5.5 mg of ester for 3 mg of retinol. Commercial oil solutions and beadlets carry the ester at a smaller concentration, and the supplier assay sets the real weight.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up
Softgel5.5 mg ester (as an oil solution of known assay)250 mg oil fillCarrier oil to fill weightShell compatibility, light and oxygen exposure, content assay of vitamin A
Capsule5.5 mg ester (as a beadlet of known assay)250 mg per capsuleCarrier and flow agent to fill weightBlend uniformity, fill weight variation, content assay of vitamin A

Tablet, liquid and gummy versions are separate projects, because the sensitivity of vitamin A to light and air needs its own protection plan in those systems.

Formulation Considerations

Label unit and basis. Reference labels state vitamin A in mg, in IU and in mcg RAE, and the factor from IU to micrograms depends on whether the vitamin A is retinol or a provitamin A carotenoid. The unit and the basis are written into the specification, and no conversion is applied between the three option sets on this page.
Upper intake level. The NIH Office of Dietary Supplements lists 3,000 mcg per day of preformed vitamin A as the tolerable upper intake level for adults aged 19 and older, which equals the 3 mg default, and notes that intakes of preformed vitamin A above the level can cause congenital birth defects [3]. The 7.5 mg and 25,000 IU options are above that level when the vitamin A is retinol, so every tier is settled in the market review before pilot work.
Long-term safety findings. A review of acute and chronic toxicity reports that osteoporosis and hip fracture are associated with preformed vitamin A intakes only twice the current recommended allowance, and that subtoxicity may be a growing concern [2]. The label statement and the amount tier for the finished product are reviewed against these findings.
Light and oxidation. Retinyl esters are sensitive to light and oxygen, so a softgel shell, an amber bottle and an antioxidant in the oil are set early, and potency through shelf life is checked in stability work.

Customization Options

Brands adjust the amount per serving, the vitamin A source, the label unit and the format.

The pilot run confirms shell compatibility and oxygen exposure in the softgel, blend uniformity of the beadlet in the capsule, and the content assay of the first units against the declared vitamin A basis.
