Glucosamine, Chondroitin & MSM

Glucosamine, chondroitin and MSM reference formula at example amounts of 1,500 mg, 1,200 mg and 500 mg per serving, developed as a three-tablet serving or a four-capsule serving.

Glucosamine, Chondroitin & MSM is a reference formula for brands building a three-active joint-nutrition product around example amounts of 1,500 mg of glucosamine, 1,200 mg of chondroitin and 500 mg of methylsulfonylmethane per serving. The reference label behind the default amounts is a three-tablet serving, and the brand sets the final amounts for its market.

Tablet is the lead format because the three actives weigh more than 3.5 g as raw materials per serving and are split over three tablets, and a four-capsule version carries smaller example amounts. The development work centers on gram-level unit weight, the glucosamine and chondroitin salts, moisture control and the label basis of each amount.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
Glucosamine1500 mg (example amount)Glucosamine hydrochloride, specified on a glucosamine basis; 1,500 mg glucosamine per servingStabilized glucosamine sulfate, with the input recalculated to 1,500 mg of glucosamine
Chondroitin1200 mg (example amount)Chondroitin sulfate sodium, specified on a chondroitin sulfate basis; 1,200 mg per servingChondroitin sulfate from another source, declared on its own basis
MSM500 mg (example amount)Methylsulfonylmethane (dimethyl sulfone), crystalline powder; specification 500 mg MSM per servingA different particle grade of the same compound

Amounts are stated as mg of glucosamine, mg of chondroitin sulfate and mg of methylsulfonylmethane. Free glucosamine is C6H13NO5, 179.17 g/mol and the hydrochloride, C6H14ClNO5, 215.63 g/mol, is 83.1% glucosamine by weight, so 1,500 mg of glucosamine needs about 1,805 mg of the salt. Chondroitin sulfate is a polymer, listed by PubChem as C13H21NO15S, 463.37 g/mol per repeating unit (PubChem CID 24766), and its content is set by the supplier assay rather than by a salt conversion. MSM is C2H6O2S, 94.14 g/mol (CID 6213).

Intended Benefits & Formula Rationale

This formula is designed for products positioned around joint nutrition. Glucosamine, chondroitin and MSM are included together so that one serving carries the three actives most often combined in joint products.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Joint nutritionChondroitinA Cochrane review of 43 randomized trials reports clinically meaningful lower pain scores with chondroitin than with placebo in studies shorter than 6 months, with a low level of evidence and high heterogeneity between trials [1]. Evidence: Mixed
Combination of glucosamine and chondroitinGlucosamine, ChondroitinA meta-analysis of 25 randomized trials found that chondroitin sulfate reduced pain intensity and improved physical function and that glucosamine sulfate reduced joint space narrowing, while their combination did not significantly improve symptoms, a result that the authors link to the limited number of combination trials [2]. Evidence: Contested

The amounts are examples, and the combination is a design choice that the brand and its market review confirm. The evidence for the combination is limited, so claims on a finished product are reviewed for each sales market before a product is developed.

Available Formats

The glucosamine, chondroitin and MSM reference formula is built for a three-tablet version and a four-capsule version; counts per serving are design choices that the brand can adjust.

Raw Material Input & Fill (Design Estimate)

Design estimates. The input is each labeled amount divided by the assumed assay of its raw material at 100%: glucosamine hydrochloride at 83.1% glucosamine (1,805 mg for 1,500 mg; 1,203 mg for 1,000 mg), chondroitin sulfate at its declared weight and MSM at its declared weight.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up
TabletPer tablet: glucosamine HCl 602 mg, chondroitin 400 mg, MSM 167 mg (1169 mg)1500 mg tablet weight331 mgCompression behavior at gram-level load, hardness, disintegration, moisture, content assay of the three actives
CapsulePer capsule: glucosamine HCl 301 mg, chondroitin 125 mg, MSM 125 mg (551 mg)600 mg per capsule49 mgFill weight variation, flow, moisture, content assay of the three actives

Powder, liquid and gummy versions are separate projects, because a gram-level load of three actives needs its own matrix, taste and stability work in those systems.

Formulation Considerations

Unit weight and count. About 3.5 g of raw material per serving sets the tablet weight and the count of three tablets a day. A brand that wants fewer units moves to smaller example amounts, and the count is fixed together with the pack.
Hygroscopic salts. Glucosamine hydrochloride and chondroitin sulfate pick up moisture, and MSM is a free-flowing crystal. Blend humidity, a coated tablet or sealed capsule and a desiccant in the bottle are set early to limit caking and softening.
Label basis of each amount. Glucosamine can be declared as glucosamine or as a salt, and chondroitin as chondroitin sulfate. The specification states the basis of each of the three amounts, and finished units are assayed on that basis.
Combination evidence. The meta-analysis in [2] did not find a significant symptom benefit for the glucosamine and chondroitin combination, and the Cochrane review in [1] rates its chondroitin evidence as low with high heterogeneity. Claims, amounts and the dose limit for each sales market are reviewed before a product is developed.

Customization Options

Brands adjust the amount of each active, the salts, the format and the add-ons.

ParameterRangeNote
Glucosamine per serving1000 or 1500 mgExamples for the brand; salt weight scales with the amount
Chondroitin per serving500, 800, 1200 or 1500 mgExamples for the brand; declared as chondroitin sulfate
MSM per serving300, 500 or 750 mgExamples for the brand
FormatTablet or capsulePowder, liquid and gummy are separate projects
Add-onsVitamin C, hyaluronic acid, turmeric extractEach added active increases the unit weight and the assay count

The pilot run confirms compression behavior and hardness at the gram-level load, capsule fill weight and flow, moisture behavior of the salts, and the content assay of glucosamine, chondroitin and MSM in the first units against the labeled amounts.
