Vitamin D

Single-ingredient vitamin D reference formula at 25 mcg (1,000 IU) per serving, developed as a softgel, tablet or capsule.

Vitamin D is a reference formula for brands building a daily, single-ingredient vitamin D product around a default amount of 25 mcg. The formula carries cholecalciferol (vitamin D3) as the only active ingredient, so the development work centers on the raw material form, low-dose uniformity and the carrier system for each format.

The 25 mcg default sits between the 10 mcg and 50 mcg options, and brands can move to any of the 10, 25, 50 or 125 mcg amounts without changing the structure of the formula. Softgel is the lead format because an oil carrier suits a fat-soluble vitamin; tablet and capsule versions use a diluted vitamin D3 premix to spread a very small active amount through the fill.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
Vitamin D25 mcg (default amount)Vitamin D3 (cholecalciferol) as an oil solution for softgels and as a dry beadlet or powder premix for tablets and capsules; specification 25 mcg (1,000 IU) per unit on a cholecalciferol basisVitamin D2 (ergocalciferol), set at the same 25 mcg per unit; D3 is the lead form because it raised serum 25(OH)D more than D2 in a meta-analysis of randomized trials [1]

Amounts are stated as mcg of vitamin D (cholecalciferol, C27H44O, 384.6 g/mol; PubChem CID 5280795). Vitamin D is not a salt or hydrate, so no salt-to-active conversion applies. Where a label uses IU, IU and mcg are stated together at 40 IU = 1 mcg, the relation given in [2]: 25 mcg = 1,000 IU.

Intended Benefits & Formula Rationale

This formula is designed for products positioned around everyday vitamin D intake and the nutritional side of bone and joint care. Vitamin D is included as the single active so that the amount, the form and the carrier can be set without interaction with other actives.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Daily vitamin D intakeVitamin D3Included as the sole active at 25 mcg (1,000 IU); the Institute of Medicine summary sets its vitamin D intake values on a serum 25-hydroxyvitamin D level of at least 20 ng/ml [3]. Evidence: Referenced
Bone and skeletal nutritionVitamin D3Positioned around bone-health nutrition because the 2011 IOM dietary reference intakes for vitamin D were derived on skeletal outcomes [3]. Evidence: Referenced
Form selection for raising serum levelsVitamin D3 rather than D2A meta-analysis of randomized trials found D3 more efficacious than D2 at raising serum 25(OH)D, with the difference lost under daily dosing [1]. Evidence: Referenced

The design keeps one active and one clear amount per unit. Brands that want a different tier adjust the premix input and retain the carrier system, and brands that want a mineral partner move to a separate combination project.

Available Formats

Three formats can be developed from this reference formula, each at one unit per serving. Pack counts in the table are opening suggestions for the brand to adjust.

Raw Material Input & Fill (Design Estimate)

Design estimates. The input is the labeled amount divided by the assumed potency of the raw material, taken at 100% assay: vitamin D3 oil or premix at 100,000 IU/g (2,500 mcg/g), so 25 mcg needs 10 mg per unit.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up

Tablet10 mg D3 premix200 mg190 mgBlend uniformity, hardness, content assay
Capsule10 mg D3 premix200 mg190 mgBlend flow, fill weight variation, content assay

Liquid and gummy versions are separate projects, because a liquid needs a different carrier system and a gummy needs a different matrix.

Formulation Considerations

Raw material form. Cholecalciferol (D3) is the lead form, and the 2012 meta-analysis of randomized trials favors it over D2 for raising serum 25(OH)D [1]. D2 remains a form option when a brand needs it, set at the same 25 mcg per unit.
Very small active amount. At 25 mcg and an assumed 10 mg of premix per unit, the active is a tiny part of a 200 mg tablet or capsule fill. Blend uniformity and fill weight variation decide whether each unit holds the labeled amount, so the premix is diluted before it reaches the main blend.
Oil carrier and softgel shell. In a softgel the vitamin is dissolved in the carrier oil, which keeps the active spread through the fill. The carrier choice also affects shell compatibility and oxidation behavior, so it is chosen with the softgel shell in the pilot run.
Amount tiers and label units. Brands can choose amounts from 10 mcg (400 IU) to 125 mcg (5,000 IU). Whichever tier is chosen, the label states mcg with IU in parentheses at 40 IU = 1 mcg [2]. Amounts above 125 mcg are handled as separate high-dose projects.

Customization Options

Brands adjust the amount per serving, the vitamin D form, the carrier and the pack format.

The pilot run confirms blend uniformity of the small vitamin D3 amount, carrier and shell compatibility for softgels, and the content assay of the first units against the labeled 25 mcg.
