Melatonin

Single-ingredient melatonin reference formula at 3 mg per serving, developed as a tablet, capsule or gummy.

Melatonin is a reference formula for brands building a single-ingredient melatonin product around a default amount of 3 mg. The formula carries melatonin as the only active ingredient, so the development work centers on distributing a milligram-level active evenly through each unit and on keeping the content in each format close to the labeled amount.

The default amount is 3 mg per serving. Tablet is the lead format, capsule is a close alternative on the same premix, and gummy covers the chewable segment.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
Melatonin3 mg (default amount)Melatonin (N-acetyl-5-methoxytryptamine) powder, synthetic, used neat or as a diluted premix; specification 3 mg melatonin per servingPre-diluted melatonin blend at the same 3 mg of melatonin, with the diluent and potency set with the supplier

Amounts are stated as mg of melatonin (C13H16N2O2, 232.28 g/mol; PubChem CID 896). Melatonin is not a salt or hydrate, so no salt-to-active conversion applies.

Intended Benefits & Formula Rationale

This formula is designed for products positioned around nighttime routines and sleep-related nutrition. Melatonin is included as the single active so that the amount and the format can be set without interaction with other actives.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Sleep-related nutritionMelatoninPositioned around sleep because a meta-analysis of randomized, placebo-controlled trials in people with primary sleep disorders found shorter sleep latency with melatonin (weighted mean difference 7.06 minutes, 95% CI 4.37 to 9.75), with effects described as modest [1]. Evidence: Referenced; the trials were in patients, not general users
Dose and format selectionMelatoninA JAMA research letter analyzed the quantity of melatonin in gummy products sold in the US [2], which is why the content assay is part of the plan for every format. Evidence: Referenced

The design keeps one active and one clear amount per unit. Brands that want a different tier change the input and keep the format structure, and brands that want a sleep blend with other actives move to a separate combination project.

Available Formats

Three formats can be developed from this reference formula, each at one unit per serving. Pack counts here are starting suggestions only.

Capsule1 capsule3 mg60 or 120 capsulesFiller, flow agentSame premix as the tablet; simple to develop
Gummy1 gummy3 mg60 gummiesPectin, sweetener, acidulant, flavor, colorChewable segment; needs a gummy matrix and content assay

Raw Material Input & Fill (Design Estimate)

Design estimates. The input is the labeled amount divided by the assumed assay of the raw material, taken at 100% melatonin, so 3 mg needs 3 mg of raw material per unit.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up
Tablet3 mg200 mg tablet weight197 mgBlend uniformity, hardness, disintegration, content assay
Capsule3 mg150 mg fill147 mgPremix uniformity, fill weight variation, content assay
Gummy3 mg3000 mg gummy weight2997 mgDosing uniformity across the depositing run, content assay after cooling and curing

Liquid and softgel versions are separate projects, because each needs a different carrier system for a small amount of active.

Formulation Considerations

Small active in a large unit. At 3 mg in a 200 mg tablet the active is 1.5% of the unit, and in a 3000 mg gummy it is 0.1%. Uniformity is the central issue in every format, so melatonin is pre-blended with part of the diluent before it enters the main blend or the gummy mass.
Content accuracy. A published analysis looked at the quantity of melatonin in gummy products sold in the US [2]. For that reason the plan fixes a content assay for first units in each format and a release limit for the labeled 3 mg.
Gummy matrix. A gummy adds heat and moisture steps that tablets do not have. The mass is cooked, the melatonin is added at a controlled temperature, and the depositing run is sampled at the start, middle and end to confirm even distribution.
Amount tiers. Amounts of 1, 3, 5 or 10 mg are the options considered. Moving from 3 mg to 1 mg or 10 mg changes the dilution step for the premix, not the unit design.

Customization Options

Brands adjust the amount per serving, the format, the premix and the flavor.

The pilot run confirms blend or mass uniformity of the 3 mg active across the batch, the content assay of the first units against the labeled amount, and, for gummies, the effect of the cooking and curing steps.
