L-Tryptophan

L-tryptophan reference formula at a default of 500 mg per serving, developed as a capsule, powder or tablet.

L-Tryptophan is a reference formula for brands building a single-ingredient essential amino acid product around 500 mg per serving. The brand sets the final amount for its market, and reference labels span 100 to 2,000 mg per serving.

Capsule is the lead format because most reference labels use it, with powder and tablet developed alongside. The work centers on a crystalline amino acid with limited water solubility and a bitter taste, where purity of the raw material, light protection and dose per unit decide the formula.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
L-Tryptophan500 mgL-tryptophan free-form powder, fermentation-derived, specified on a declared-weight basis; 500 mg per servingA different purity grade, with the impurity limits stated on the specification

The amount is stated as mg of L-tryptophan. L-tryptophan is C11H12N2O2, 204.22 g/mol (PubChem CID 6305); the free form has no salt or hydrate conversion.

Intended Benefits & Formula Rationale

This formula is designed for products positioned around evening and sleep nutrition. L-tryptophan is included as the single ingredient so that the concept stays on one essential amino acid.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Sleep continuityL-TryptophanA systematic review and meta-analysis of tryptophan supplementation reports that supplementation shortened wake after sleep onset (SMD −1.08; 95% CI −1.89 to −0.28), with a shorter wake after sleep onset at 1 g or more per day than at less than 1 g [1]. Evidence: Referenced
Other sleep componentsL-TryptophanThe same review reports that tryptophan supplementation did not affect other sleep components, and its meta-analysis of the effect on wake after sleep onset rests on four articles [1]. Evidence: Mixed

The amount is a design choice. The review in [1] covers several doses and does not test this formula, so any claim on a finished product is reviewed for each market.

Available Formats

Reference labels are mostly capsules, with powders and tablets behind them; these three formats are developed here.

Raw Material Input & Fill (Design Estimate)

Design estimates. L-tryptophan is assumed at 100% of its declared weight, so 500 mg per serving needs 500 mg of raw material; the supplier’s assay is applied once the lot is chosen.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up

Tablet500 mg650 mg tablet weight150 mgCompression, hardness, coating, disintegration

Liquid and gummy are separate projects, because the limited solubility of tryptophan in water changes the formulation work in those systems.

Formulation Considerations

Raw material purity. Tryptophan quality depends on the fermentation and purification process, and the specification sets impurity limits and the identity tests. Supplier qualification is part of the plan before any batch.
Solubility. L-tryptophan dissolves slowly in cold water, so a drink mix settles to the bottom. Particle size, a dispersing agent and mixing instructions are tested for the powder format.
Light and oxidation. The indole ring is sensitive to light and oxygen over time, which can darken the powder. Opaque packaging and a stability assay are part of the plan.
Bitterness. The free amino acid is bitter. Capsules and coated tablets hide it, and the powder format needs a sweetener and acid system.

Customization Options

Brands adjust the amount, the format and the add-ons.
