Coenzyme Q10

Single-ingredient coenzyme Q10 reference formula at 100 mg per serving, developed as a softgel, capsule or tablet.

Coenzyme Q10 is a reference formula for brands building a single-ingredient CoQ10 product around a default amount of 100 mg. The formula carries ubidecarenone (coenzyme Q10) as the only active ingredient, so the development work centers on how a hydrophobic, high-molecular-weight active is carried in the fill and how it is delivered in each format.

The default amount is 100 mg per serving. Softgel is the lead format because an oil system carries the active, while capsule and tablet versions use powder CoQ10 with their own flow and compression work.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
Coenzyme Q10100 mg (default amount)Ubidecarenone (coenzyme Q10) crystalline powder, suspended in an oil carrier for softgels and used as a powder in capsules and tablets; specification 100 mg coenzyme Q10 per servingUbiquinol (the hydroquinone form) as a separate raw material, with the input calculated from its own molecular weight and set with the supplier; pre-solubilized CoQ10 raw material at the same 100 mg of coenzyme Q10

Amounts are stated as mg of coenzyme Q10 (ubidecarenone, C59H90O4, 863.3 g/mol, from the verified molecular weight record). CoQ10 is not a salt or hydrate, so no salt-to-active conversion applies to the lead form.

Intended Benefits & Formula Rationale

This formula is designed for products positioned around heart and circulatory nutrition and daily CoQ10 intake. Coenzyme Q10 is included as the single active so that the amount, the carrier and the format can be set without interaction with other actives.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Cellular energy nutritionCoenzyme Q10Included for its role in cellular bioenergetics and as an antioxidant [2][3]. Evidence: Referenced
Heart and circulatory nutritionCoenzyme Q10Positioned around heart and circulatory nutrition because CoQ10 has been studied as an adjunct in chronic heart failure in a randomized, double-blind trial [3] and examined in a systematic review of systematic reviews in cardiovascular disease [1]. Evidence: Referenced; the studies were in patient populations
Delivery systemCoenzyme Q10 in an oil carrierAbsorption of dietary CoQ10 is slow and limited, and solubilized supplement formulations show enhanced bioavailability [2]. Evidence: Referenced

The design sets one active and one clear amount per unit and puts the effort into the carrier system. Brands that want a different tier change the input and the fill size, and brands that want a ubiquinol start a separate raw material discussion.

Available Formats

Three formats can be developed from this reference formula, each at one unit per serving. The brand picks the final count per bottle.

FormatUnitPer ServingPack SizeOther Ingredients (typical)Suitability

Tablet1 tablet100 mg60 or 90 tabletsCellulose, disintegrant, flow agent, magnesium stearateCompressed unit; needs a granulated blend

Raw Material Input & Fill (Design Estimate)

Design estimates. The input is the labeled amount divided by the assumed assay of the raw material, taken at 100% coenzyme Q10, so 100 mg needs 100 mg of raw material per unit.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up
Softgel100 mg400 mg fill300 mg oil systemSuspension uniformity in the fill, fill weight variation, content assay
Capsule100 mg300 mg fill200 mgPowder flow, fill weight variation, content assay
Tablet100 mg400 mg tablet weight300 mgBlend uniformity, hardness, disintegration, content assay

Liquid and gummy versions are separate projects, because a gummy matrix and a liquid carrier each need their own system for an oil-soluble active.

Formulation Considerations

Hydrophobic active and absorption. CoQ10 is hydrophobic with a molecular weight of 863.3 g/mol, and its dietary absorption is slow and limited, while solubilized formulations show enhanced bioavailability [2]. This is the reason the softgel with an oil system is the lead format.
Suspension in the softgel fill. In the oil fill the active is a suspended powder, so particle size, viscosity and suspending agent (lecithin appears in the reference formula) decide whether the 100 mg stays evenly spread through the 400 mg fill. Settling is checked during fill preparation and shell sealing.
Shell choice. The reference formula uses a modified starch and carrageenan shell, which is a plant-based alternative to gelatin. The shell type is chosen with the brand’s label requirements and is tested for compatibility with the oil fill.
Amount tiers. The options considered run from 30 mg to 200 mg. A 200 mg serving doubles the raw material mass in the same fill, so it moves to a larger softgel or to two units per serving.

Customization Options

Brands adjust the amount per serving, the CoQ10 raw material, the carrier and the shell.

The pilot run confirms suspension stability of the CoQ10 in the oil fill, softgel shell compatibility, and the content assay of the first units against the labeled 100 mg.
