Astaxanthin

Single-ingredient astaxanthin reference formula at 12 mg per serving, developed as a softgel or capsule.

Astaxanthin is a reference formula for brands building a single-ingredient carotenoid product at a default amount of 12 mg of astaxanthin per serving. Astaxanthin is the only active ingredient, so the development work centers on a lipophilic, oxygen-sensitive pigment: the natural microalgae source, the oil that carries it, and the shell or bottle that holds its color and potency through shelf life.

Softgel is the lead format because astaxanthin is fat soluble and is delivered in an oil fill, while a capsule version uses an oil-based or beadlet form. Options of 4, 6 and 10 mg sit beside the default, and the amount is a decision for the brand and its market review.

Ingredient Composition

IngredientPer ServingRaw Material FormAlternative Form (same amount of active)
Astaxanthin12 mg (default amount)Natural astaxanthin from Haematococcus pluvialis microalgae as an oleoresin, specified on astaxanthin content; 12 mg astaxanthin per servingA beadlet or powder form of the same source for capsules, with the input recalculated to 12 mg of astaxanthin

Amounts are stated as mg of astaxanthin (C40H52O4, 596.8 g/mol; PubChem CID 5281224). Microalgae astaxanthin occurs mainly as esters, and the assay basis, free astaxanthin or total esters expressed as astaxanthin, is stated in the specification with the supplier. Synthetic astaxanthin is a different raw material and is not part of this specification.

Intended Benefits & Formula Rationale

This formula is designed for products positioned around antioxidant nutrition and healthy aging. Astaxanthin is included as the single active so that the source, the amount and the oil system can be set without interaction with other actives.

Intended Benefit DirectionKey IngredientsFormulation Rationale
Antioxidant nutritionAstaxanthinA meta-analysis of 12 randomized trials with 380 participants found that astaxanthin significantly reduced blood malondialdehyde compared with placebo (standardized mean difference -0.95), and described the effects on oxidative stress and inflammation biomarkers as mild [1]. Evidence: Referenced
Skin nutritionAstaxanthinA systematic review and meta-analysis found that oral astaxanthin restored skin moisture and improved elasticity versus placebo, and did not significantly decrease wrinkle depth [2]. Evidence: Mixed

The amount is one stated tier. The reference amounts range from 4 to 12 mg per unit, and the trials in [1] and [2] differ in dose and duration, so amount, claims and target market are brand decisions reviewed for each sales market.

Available Formats

The astaxanthin reference formula is built for a softgel version and a capsule version; counts per serving are design choices that the brand can adjust.

Raw Material Input & Fill (Design Estimate)

Design estimates. The input is the astaxanthin amount divided by the astaxanthin content of the raw material at an assumed assay: a 10% astaxanthin oleoresin gives 120 mg of oleoresin per 12 mg of astaxanthin, and a 2% beadlet would give 600 mg.

FormatRaw Material Input per UnitDesign FillBalance for ExcipientsVerification Before Scale-Up

Capsule120 mg of a 10% powder or beadlet equivalent250 mg per capsule130 mgBlend uniformity, fill weight variation, astaxanthin assay

Tablet, liquid and gummy versions are separate projects, because an oxygen-sensitive pigment needs its own protection plan in those systems.

Formulation Considerations

Oxygen and light. Astaxanthin is a deeply colored carotenoid that oxidizes with air, heat and light. Mixed tocopherols and rosemary extract in the oil, nitrogen flushing at fill, opaque shells or amber bottles and potency through shelf life are set in stability work.
Oleoresin assay and color. The oleoresin is viscous and dark red, and its astaxanthin share sets the fill. Color uniformity between softgels and staining of equipment are checked in the pilot run.
Natural or synthetic. Reference labels often state natural astaxanthin from microalgae, while the assay basis, free or esterified, is part of the specification. The source is declared on the label and the formula, and a switch to another source is a new product.
Evidence and tier. The meta-analyses in [1] and [2] cover varied doses and durations, and the effects are mild or partial. Which tier a brand picks follows its sales market, whose dose limit is checked before development starts.

Customization Options

Brands adjust the amount per serving, the astaxanthin source and assay, the format and the oil system.
